Abstract: A mucosite intestinal (MI) causada pelas terapias anticâncer, caracteriza-se por um processo inflamatório da mucosa e pode ocorrer entre 50% e 80% dos pacientes oncológicos tratados com 5-Fluorouracil (5-FU), quimioterápico comumente utilizado. Apesar da elevada incidência entre os pacientes, não existem terapêuticas aprovadas para a prevenção e/ou cura da MI. Diversos estudos têm demonstrado capacidade antioxidante e anti-inflamatória do açaí (Euterpe oleracea Martius), fruto rico em compostos fenólicos, sendo o principal, as antocianinas e cada vez mais consumido pela população mundial. Diante disso, o objetivo deste estudo foi avaliar o efeito da dieta suplementada com polpa de açaí 2% sobre a quantificação da Interleucina-10 (IL-10) de camundongos portadores de MI induzida por 5-FU. Camundongos fêmeas da linhagem BALB/c foram divididos em quatro grupos experimentais: Controle (C): recebeu dieta controle AIN-93M e não foi induzida mucosite; Açaí (AÇ): recebeu dieta suplementada com polpa de açaí 2% e não foi induzida mucosite; Mucosite (M): recebeu dieta controle e foi induzida mucosite por 5-FU; Açaí + Mucosite (AÇM): recebeu dieta suplementada com polpa de açaí 2% e foi induzida mucosite. Foram inseridas as respectivas dietas por um período de quatorze dias consecutivos. No décimo quinto dia foi realizada a indução da MI nos grupos M e AÇM. Os grupos C e AÇ receberam injeção intraperitoneal (ip) de tampão fosfatosalino (PBS - Phosphate-Buffered Saline) estéril. Executou-se a eutanásia três (D3) e sete (D7) dias após a indução da mucosite, e, em seguida, foi dissecado o duodeno e íleo. A quantificação da concentração IL-10 nos tecidos coletados dos animais foi realizada por meio do ensaio imunoenzimático do tipo ELISA. Observou-se que a concentração de IL-10 foi negativa em todos os grupos, ou seja, as concentrações estão abaixo do nível de sensibilidade do teste e isso indica que a quantidade de IL-10 no duodeno e íleo é pouco significativa. Além disso, o consumo de açaí, não demonstrou um aumento na produção de IL-10. O sistema imune é muito complexo e integrativo, e, por isso, não se pode limitar a observação de um fenômeno como a inflamação pela perspectiva de apenas um mediador inflamatório. É necessário a realização de mais estudos que melhor esclareçam as vias de atuação do açaí no possível efeito anti-inflamatório na MI experimental, possibilitando sua utilização como estratégia dietética preventiva e/ou terapêutica para a doença.
metadata.dc.description.abstracten: Intestinal mucositis (IM) caused by anticancer therapies is characterized by an inflammatory process of the mucosa and can occur in 50% to 80% of patients with cancer treated with 5-Fluorouracil (5-FU), a commonly used chemotherapy. Despite the high incidence among patients, there aren’t approved therapies for the prevention and/or cure of IM. Several studies have demonstrated the antioxidant and anti-inflammatory capacity of açaí (Euterpe oleracea Martius), a fruit rich in phenolic compounds, being the main, the anthocyanins and increasingly consumed by the world population. Therefore, the aim of this study was to evaluate the effect of the diet supplemented with 2% açaí pulp on the quantifying of Interleukin-10 (IL-10) in mice bearing 5-FU induced IM. Female BALB/c mice were divided into four experimental groups: Control (C): received a control diet AIN-93M and no mucositis was induced; Açaí (AÇ): received a supplemented diet with pulp of açaí 2% and no mucositis was induced; Mucositis (M): received a control diet and mucositis was induced by 5-FU; Açaí + Mucositis (AÇM): received a supplemented diet with pulp of açaí 2% and mucositis was induced. The respective diets were entered for a period of fourteen consecutive days. On the fifteenth day, IM induction was performed in groups M and AÇM. Groups C and AÇ received an intraperitoneal (ip) injection of PBS (Phosphate-Buffered Saline). Euthanasia was performed three (D3) and seven (D7) days after mucositis induction, and then the duodenum and ileum were dissected. The quantification of the IL-10 concentration in the tissues collected from the animals was performed using an enzyme-linked immunosorbent assay of the ELISA type. It was observed that the concentration of IL-10 was negative in all groups, that is, the concentrations are below the sensitivity level of the test, and this indicates that the amount of IL-10 in the duodenum and ileum isn’t very significant. Furthermore, the consumption of açaí didn’t show an increase in the production of IL-10. The immune system is very complex and integrative, and, therefore, it cannot limit the observation of a phenomenon such as an inflammation from the perspective of just one inflammatory mediator. It`s necessary to carry out more studies to better clarify the ways in which açaí acts in the possible anti-inflammatory effect in experimental IM, enabling its use as a preventive and/or therapeutic dietary strategy for the disease.
